De behoefte aan een vaccin tegen HIV is onmiskenbaar, maar de ontwikkeling van een goed werkend vaccin heeft veel voeten in de aarde. De HIV-envelopglycoproteïnen, te vinden op het virale membraan, zijn de enige virale proteïnen die aan de buitenkant van de virusdeeltjes zitten. Deze staan dus het eerst bloot aan een vaccin.
In het eerste deel van dit proefschrift beschrijft het onderzoek naar de functionaliteit en eigenschappen van de hiv-1-envelopglycoproteïnen. Het tweede deel van het proefschrift beschrijft het ontwerp van modificaties aan de envelopglycoproteïnen, die ervoor moeten zorgen dat de eigenschappen als vaccinatieantigenen worden verbeterd.
